Overview

CocH3-Fc(M3) is a long-acting, engineered fusion protein designed for the treatment of cocaine addiction. It consists of a highly efficient mutant human butyrylcholinesterase (CocH3) fused to an engineered fragment crystallizable (Fc) domain, specifically the M3 variant, to extend its biological half-life. The enzyme rapidly metabolizes and inactivates cocaine in the bloodstream by hydrolyzing it into biologically inactive metabolites, thereby preventing both its physiological and toxic effects. Unlike traditional pharmacological approaches that target brain receptors or transporters, CocH3-Fc(M3) acts peripherally without affecting normal brain function or other critical organs. Preclinical studies demonstrate that a single dose can block cocaine’s effects for up to two weeks or longer in animal models, making it a promising candidate for relapse prevention and overdose protection during addiction therapy[1][2][4][5].
