Col-M@AuNCs + doxorubicin is an experimental multifunctional biomimetic nanoplatform designed for the treatment of pancreatic ductal adenocarcinoma (PDAC). The system consists of gold nanoclusters (AuNCs) loaded with the chemotherapeutic agent doxorubicin, functionalized with the enzyme collagenase (Col), and encapsulated within a biomimetic macrophage membrane (M). This platform is specifically engineered to overcome the dense extracellular matrix (ECM) of pancreatic tumors, where collagen acts as a significant physical barrier to drug penetration. The collagenase component degrades collagen fibers within the tumor stroma, facilitating deeper intratumoral diffusion of the nanoparticles and the doxorubicin payload. Furthermore, the gold nanoclusters enable synergistic photothermal therapy (PTT) and photodynamic therapy (PDT) upon near-infrared irradiation, while also serving as a contrast agent for computed tomography (CT) imaging. The macrophage membrane coating provides immune evasion and tumor-homing capabilities, enhancing the overall therapeutic efficacy and targeting of the platform.
