Overview

Colchicine + paracetamol is a combination treatment regimen evaluated in the Phase 3 COLCHICOVID clinical trial conducted by the Instituto de Investigación Marqués de Valdecilla (IDIVAL) in Spain. This regimen combines colchicine, a small molecule anti-inflammatory drug traditionally used for gout, with paracetamol (acetaminophen), a common analgesic and antipyretic used for symptomatic relief. Colchicine acts by binding to tubulin and inhibiting microtubule polymerization, which interferes with the activation of the NLRP3 inflammasome and the subsequent release of pro-inflammatory cytokines such as IL-1β and IL-18. In the context of COVID-19, this combination is investigated for its potential to mitigate the hyperinflammatory response and prevent severe pulmonary complications and mortality in high-risk, non-hospitalized elderly patients (over 60 years old) with comorbidities.
