Overview
Colistimethate + zidovudine is an investigational combination therapy being developed primarily to address infections caused by multidrug-resistant Gram-negative bacteria, such as Escherichia coli and carbapenem-resistant Klebsiella pneumoniae. Colistimethate (also known as colistin) is a polymyxin antibiotic that acts by disrupting the bacterial cell membrane, leading to cell death. Zidovudine (azidothymidine, AZT) is a nucleoside reverse transcriptase inhibitor originally developed for HIV/AIDS treatment; in this context, it acts synergistically with colistin to enhance antibacterial activity against resistant strains. The combination has demonstrated significant synergistic bactericidal effects in preclinical models and in vitro studies, reducing bacterial burden more effectively than either agent alone[1][3][5][8].
