COM701 + nivolumab

COM701 + nivolumab is a combination of two immunotherapy drugs under investigation for the treatment of advanced solid tumors. COM701 is a first-in-class humanized IgG4 monoclonal antibody that targets and blocks poliovirus receptor related immunoglobulin domain containing (PVRIG), an immune checkpoint protein. By inhibiting PVRIG’s interaction with its ligand PVRL2, COM701 enhances activation of T cells and natural killer (NK) cells, potentially leading to antitumor effects[1][5][8]. Nivolumab is a well-established monoclonal antibody targeting programmed cell death protein 1 (PD-1), blocking its interaction with PD-L1/PD-L2 to promote immune-mediated tumor cell killing. The rationale for combining these agents is to simultaneously block multiple inhibitory pathways in the DNAM axis—specifically PVRIG and PD-1—to enhance antitumor immunity in patients who have exhausted standard treatments[5][8]. This combination has shown preliminary safety and antitumor activity in early-phase clinical trials across several solid tumor types including metastatic breast cancer, microsatellite stable colorectal cancer (MSS-CRC), ovarian cancer, endometrial cancer, non-small cell lung cancer (NSCLC), and others[3][5].
