Overview
Complete Freund's Adjuvant is a **non-specific, variable-composition immunologic adjuvant preparation**, rather than a single chemically defined active pharmaceutical ingredient. Common preparations are water-in-oil systems containing mineral or paraffin oil, a surfactant such as mannide monooleate, and heat-killed mycobacteria; commercial products can differ in the mycobacterial species and concentration. The oil phase can create a local antigen depot, while mycobacterial material promotes local innate immune-cell recruitment and inflammatory activation. Although primarily used as a research immunization reagent, CFA was evaluated by the Canberra Hospital Medical Oncology Department in an Australian Phase I study involving intratumoral CFA administration in patients with refractory or relapsed solid tumours; the protocol also included intravenous pembrolizumab.
