Compound 7b is a preclinical triazolothiadiazine derivative small molecule originally investigated as a nonsteroidal anti-inflammatory drug (NSAID). It has demonstrated potent anti-cancer activity against hepatocellular carcinoma (HCC) by inducing oxidative stress, which subsequently activates the c-Jun N-terminal kinase (JNK) signaling pathway, specifically involving ASK1, MKK7, and c-Jun proteins. In preclinical studies, Compound 7b was identified as the most effective lead among 32 triazolothiadiazine derivatives, inducing G2/M phase cell cycle arrest and apoptosis in HCC cells. It has also been shown to inhibit HCC cell migration and reduce liver cancer stem cell populations, both as a monotherapy and in combination with sorafenib. Preclinical animal models have demonstrated that treatment with Compound 7b significantly decreases tumor volume and prolongs disease-free survival.
