Compound 7d is a potent, orally bioavailable, type I FMS-like tyrosine kinase 3 (FLT3) inhibitor developed by Array BioPharma. It was identified as a lead compound for the treatment of FLT3-driven cancers, such as acute myeloid leukemia (AML). Unlike type II inhibitors like quizartinib, Compound 7d is a type I inhibitor, meaning it targets both the wild-type FLT3 enzyme and common resistance-conferring activation loop mutations, such as D835Y. In preclinical oncology models, Compound 7d has demonstrated the ability to produce a longer-lived inhibition of FLT3 phosphorylation compared to first-generation inhibitors, suggesting potential for improved clinical durability in patients with FLT3-mutated malignancies.
