Compound 7f is a potent, orally bioavailable small molecule antagonist targeting the endosomal Toll-like receptors 7, 8, and 9 (TLR7/8/9). Developed by Bristol Myers Squibb in collaboration with the Biocon Bristol Myers Squibb Research Center, this compound was designed to address autoimmune disorders, such as systemic lupus erythematosus and psoriasis, where overactivation of endosomal TLRs is a key driver of pathology. Preclinical studies published in 2020 highlighted its excellent on-target potency for human TLR7, TLR8, and TLR9, alongside high selectivity against other members of the TLR family. Compound 7f demonstrated balanced activity in mouse systems and showed therapeutic efficacy in animal models of lupus and psoriasis. Currently, it is recognized as a research tool compound and has not progressed into clinical development.
