Overview

Compound B (CpdB) is an orally bioavailable small molecule inhibitor of Monoacylglycerol O-acyltransferase 2 (MGAT2) developed by Takeda Pharmaceutical. MGAT2 is a critical enzyme expressed in the small intestine that is primarily responsible for the re-synthesis of triglycerides from dietary fats. By blocking MGAT2 activity, Compound B reduces postprandial plasma triglyceride elevations and enhances the release of anorectic gut hormones, specifically peptide tyrosine-tyrosine (PYY) and glucagon-like peptide-1 (GLP-1), which modulate satiety and glucose metabolism. In preclinical mouse models of high-fat diet-induced obesity and type 2 diabetes (ob/ob mice), Compound B has demonstrated efficacy in suppressing food intake, reducing body weight gain, and improving metabolic disorders, including lowering glycated hemoglobin levels. The drug is part of a series of aryl-sulfonamide derivatives and represents a potential therapeutic approach for managing obesity, type 2 diabetes, and dyslipidemia.
