**Conatumumab + doxorubicin** is a combination of a fully human monoclonal agonist antibody (conatumumab) targeting death receptor 5 (DR5) and the anthracycline chemotherapeutic agent doxorubicin. - **Conatumumab** induces apoptosis in sensitive tumor cells by activating DR5. - **Doxorubicin** intercalates into DNA and inhibits topoisomerase II, leading to DNA damage and cell death. This combination was evaluated for efficacy and safety in metastatic or locally advanced/resectable soft-tissue sarcomas as first-line therapy, but clinical studies showed that adding conatumumab to doxorubicin did not significantly improve disease control compared to doxorubicin alone, though the safety profile was acceptable[3][4]. Conatumumab was originally developed by Amgen. Doxorubicin is manufactured and marketed under various brand names by multiple pharmaceutical companies[3][5].
