Overview

**Conatumumab + gemcitabine** is a combination therapy under investigation primarily for the treatment of pancreatic cancer. Conatumumab is a fully human monoclonal antibody that acts as an agonist of death receptor 5 (DR5, also known as TRAIL receptor 2), inducing apoptosis (programmed cell death) in tumor cells. Gemcitabine is a nucleoside analog and antimetabolite used as a chemotherapeutic agent; it inhibits DNA synthesis, leading to cell death. The combination is designed to target and induce apoptosis in cancer cells through DR5 activation (conatumumab) while also disrupting DNA synthesis (gemcitabine). This dual action may result in enhanced anti-tumor efficacy. Clinical trials, including phase 2 studies, have evaluated this combination in patients with metastatic pancreatic cancer, showing moderate activity but not a clear survival benefit over standard chemotherapy[1][2][5].
