Copanlisib + trastuzumab is an investigational combination therapy consisting of copanlisib, a pan-class I phosphoinositide-3 kinase (PI3K) inhibitor, and trastuzumab, a monoclonal antibody targeting the human epidermal growth factor receptor 2 (HER2). Copanlisib inhibits PI3K signaling, which is implicated in resistance to HER2-targeted therapies. Trastuzumab binds to HER2 and blocks downstream signaling pathways that promote tumor cell proliferation. This combination has been studied primarily in patients with advanced or metastatic HER2-positive breast cancer who have progressed after prior lines of anti-HER2 therapy. Preclinical studies demonstrated synergistic activity between these agents in models resistant to HER2 therapy[1][3]. Clinical trials have shown that the combination can be safely administered with manageable toxicity and preliminary evidence of disease stabilization[1][2].
