Overview

Copper + dimercaptosuccinic acid + hyaluronic acid (Cu-DMSA-HA) is an experimental nanocatalytic therapeutic agent formulated as a nanoparticle for the treatment of non-small cell lung cancer (NSCLC). The nanoparticle is composed of a copper core coordinated with dimercaptosuccinic acid (DMSA) and surface-modified with hyaluronic acid (HA). The HA moiety serves as a targeting ligand that specifically recognizes and binds to CD44 receptors, which are frequently overexpressed on the surface of NSCLC cells, thereby enhancing tumor-specific accumulation and cellular uptake via receptor-mediated endocytosis. Once internalized, the nanoparticles deplete intracellular glutathione (GSH) and catalyze a Fenton-like reaction to generate high levels of reactive oxygen species (ROS). This oxidative stress triggers ferroptosis, downregulates glutathione peroxidase 4 (GPX4) expression, and inhibits DNA replication, ultimately suppressing tumor progression and overcoming conventional drug resistance.
