corticosteroid + colchicine

This is a combination therapy consisting of a corticosteroid and colchicine. Corticosteroids are synthetic analogs of endogenous glucocorticoids that exert potent anti-inflammatory and immunosuppressive effects by modulating gene expression, inhibiting multiple inflammatory cytokines, and suppressing leukocyte migration. Colchicine is an alkaloid that disrupts microtubule polymerization in neutrophils, thereby reducing inflammation associated with crystal-induced diseases such as gout. The combination may be used off-label for certain inflammatory conditions where both rapid suppression of inflammation (via corticosteroids) and inhibition of neutrophil-mediated processes (via colchicine) are desired. There is limited evidence for the use of this specific combination; one study evaluated prednisone (a corticosteroid) plus colchicine in primary sclerosing cholangitis but did not find significant long-term benefit[6]. Both agents are individually approved for various indications including gout flares.
