Cosibelimab + olafertinib is a combination therapy being developed by Checkpoint Therapeutics for the treatment of non-small cell lung cancer (NSCLC). Cosibelimab (CK-301) is a high-affinity, fully human IgG1 monoclonal antibody that targets programmed death-ligand 1 (PD-L1). It blocks the interaction of PD-L1 with its receptors, PD-1 and B7.1, thereby restoring anti-tumor T-cell responses. Uniquely, cosibelimab features a functional Fc region designed to potentially induce antibody-dependent cellular cytotoxicity (ADCC), providing an additional mechanism of tumor cell clearance. Olafertinib (CK-101) is an oral, third-generation, irreversible small-molecule tyrosine kinase inhibitor (TKI) of the epidermal growth factor receptor (EGFR). It is designed to selectively target EGFR-sensitizing mutations and the T790M resistance mutation while sparing wild-type EGFR, which helps minimize off-target toxicities. The combination of these two agents is intended to provide a synergistic effect by simultaneously inhibiting oncogenic driver signaling and overcoming immune evasion in EGFR-mutated NSCLC.
