COTI-2 + cisplatin

COTI-2 + cisplatin is a **combination therapy** composed of the investigational small molecule COTI-2 with the chemotherapy agent cisplatin. COTI-2 is a third-generation thiosemicarbazone derivative that targets and restores the function of mutant p53 proteins, induces apoptosis, and modulates the PI3K/AKT/mTOR pathway and cellular metabolism[1][2][4][5]. Cisplatin is a well-established DNA-damaging chemotherapeutic agent used in diverse solid tumors. Preclinical studies show that when used together, COTI-2 and cisplatin exhibit strong synergistic anti-tumor effects and are well-tolerated in animal models, particularly in head and neck squamous cell carcinoma (HNSCC), non-small cell lung cancer (NSCLC), and ovarian cancer[1][3][5]. The combination acts by enhancing DNA damage and replication stress responses, leading to increased cancer cell apoptosis and/or senescence, including in tumor cells resistant to cisplatin alone[3][5].
