CoV-RBD-scNP-001 + 3M-052-AF is a **combination vaccine candidate** targeting SARS-CoV-2 and related betacoronaviruses. The vaccine consists of a recombinant **receptor binding domain (RBD)** of the SARS-CoV-2 spike protein engineered onto a **ferritin-based nanoparticle scaffold** (scNP for sortase-conjugated nanoparticle), and is adjuvanted with **3M-052-AF**, a synthetic **Toll-like receptor 7/8 (TLR7/8) agonist** formulated in an aqueous solution. This combination is designed to induce strong and broad **neutralizing antibody responses** as well as T cell–mediated immunity. Studies in non-human primates and mice show that RBD-scNP + 3M-052-AF elicits high titers of cross-neutralizing antibodies against SARS-CoV-2, its major variants (including Beta, Delta, and Omicron), SARS-CoV-1, and related bat coronaviruses, and provides robust protection from both upper and lower respiratory tract infection. The 3M-052-AF adjuvant boosts the immunogenicity compared to formulations lacking this adjuvant[2][3][4][5][6][7][9].
