CoV2 preS dTM + AF03 is a recombinant protein subunit vaccine candidate developed by Sanofi for the prevention of COVID-19. The vaccine consists of a stabilized, soluble version of the SARS-CoV-2 spike protein (preS dTM), which has been modified by deleting the transmembrane domain and using a pre-fusion stabilized conformation. This antigen is produced using Sanofi's proprietary baculovirus expression system. The formulation is adjuvanted with AF03, a squalene-based oil-in-water emulsion developed by Sanofi to enhance the immune response. During early clinical development (Phase 1/2), this formulation was evaluated alongside an AS03-adjuvanted version (developed in partnership with GSK). While the AS03-adjuvanted version (VidPrevtyn) proceeded to Phase 3 trials and regulatory approval, the AF03-adjuvanted version was not pursued for further late-stage development.
