CPI-006 + ciforadenant

CPI-006 + ciforadenant is a combination therapy under clinical investigation for the treatment of advanced cancers. CPI-006 is a humanized monoclonal antibody that targets CD73, an ectonucleotidase involved in the conversion of AMP to adenosine, thereby blocking adenosine production and modulating immune cell activity. It exhibits immunomodulatory effects by activating B cells and enhancing antigen presentation, as well as inhibiting CD73 enzyme activity without internalization[1][4][6]. Ciforadenant (also known as CPI-444) is an oral small molecule inhibitor of the adenosine A2A receptor, designed to block the immunosuppressive effects of adenosine within the tumor microenvironment and restore anti-tumor immune responses[1][8]. The combination aims to synergistically enhance anti-tumor immunity by both preventing adenosine generation (via CD73 inhibition) and blocking its action on immune cells (via A2A receptor antagonism).
