Drug intelligence / Profile preview
CPX-351 (Vyxeos) is a liposomal encapsulation of the anthracycline daunorubicin and the antimetabolite cytarabine in a fixed 1:5 molar ratio. This formulation is designed to optimize the synergistic delivery of the two agents to leukemia cells while minimizing systemic toxicity. Daunorubicin acts by intercalating into DNA and inhibiting topoisomerase II, while cytarabine is a pyrimidine analog that inhibits DNA polymerase and incorporates into DNA, leading to chain termination. It was developed by Celator Pharmaceuticals (now Jazz Pharmaceuticals) and is primarily indicated for the treatment of adults with newly-diagnosed therapy-related acute myeloid leukemia (t-AML) or AML with myelodysplasia-related changes (AML-MRC).
