This treatment protocol is a multi-component regimen designed for patients with high-risk FLT3-mutated acute myeloid leukemia (AML). It integrates induction and consolidation therapy using CPX-351 (a liposomal combination of daunorubicin and cytarabine) and midostaurin (a FLT3 inhibitor) to achieve remission. Following this, patients undergo a conditioning regimen consisting of busulfan, melphalan, and fludarabine, followed by a CD34+ selected allogeneic stem cell transplant. The CD34+ selection process involves the enrichment of hematopoietic stem cells and the depletion of T-cells from the donor graft, a technique intended to minimize the risk of graft-versus-host disease (GVHD) while maintaining the graft-versus-leukemia effect. This specific pilot study is led by Guenther Koehne.
