CRB-601 + anti-PD-(L)1 antibody is a combination investigational therapy under clinical evaluation for advanced solid tumors. CRB-601 is a monoclonal antibody that targets and blocks the integrin αvβ8, a protein that activates latent transforming growth factor-beta (TGF-β) in the tumor microenvironment. By inhibiting integrin αvβ8, CRB-601 prevents the activation of TGF-β, a pathway that tumors exploit to suppress immune responses and create immunosuppressive barriers. This enhances immune cell infiltration and anti-tumor activity. Anti-PD-(L)1 antibodies are immune checkpoint inhibitors that block the programmed death-1 (PD-1) or its ligand PD-L1, removing inhibitory signals on T cells to promote anti-tumor immunity. The combination is designed to synergistically restore and amplify anti-tumor immune responses, overcome tumor immune resistance, and improve responses to immunotherapy in patients whose cancers are otherwise resistant to checkpoint inhibition alone. Corbus Pharmaceuticals is developing CRB-601 and is conducting ongoing phase 1/2 clinical trials combining CRB-601 with anti-PD-(L)1 antibodies in multiple advanced solid tumor types[1][2][3][4][5][6].
