Overview

CRB-701 + anti-PD-1 is a combination therapy comprising CRB-701, a next-generation antibody-drug conjugate (ADC) targeting Nectin-4, and an anti-PD-1 checkpoint inhibitor, specifically pembrolizumab. CRB-701 (also known as SYS6002) utilizes a site-specific, cleavable linker to attach a monomethyl auristatin E (MMAE) cytotoxic payload to a Nectin-4-directed antibody with a drug-antibody ratio (DAR) of 2. This combination aims to leverage the direct tumor-killing effects of the ADC and the immune-potentiating effects of PD-1 blockade. It is currently being evaluated in Phase 1/2 clinical trials for advanced solid tumors, including head and neck squamous cell carcinoma (HNSCC), cervical cancer, and metastatic urothelial cancer.
