Overview

This is an investigational combination regimen comprised of five active agents: crenolanib, fludarabine, cytarabine, granulocyte colony-stimulating factor, and idarubicin. The regimen is under investigation for the treatment of acute myeloid leukemia (AML), particularly in cases with relapsed or refractory FLT3-mutated disease and in pediatric AML. - **Crenolanib** is an oral, type I, small molecule inhibitor targeting FLT3, with activity against both FLT3-ITD and FLT3-TKD mutations, working by inhibiting FLT3-mediated signaling, cell proliferation, and survival in leukemic blasts. - **Fludarabine** is a purine analog that inhibits DNA synthesis and induces apoptosis in rapidly dividing cells. - **Cytarabine** is a nucleoside analog that blocks DNA synthesis, commonly used as a backbone of AML chemotherapy. - **Granulocyte colony-stimulating factor (G-CSF)** is a cytokine that stimulates the production and differentiation of neutrophilic granulocytes and may be used to "prime" leukemic cells, increasing their sensitivity to cytotoxic agents. - **Idarubicin** is an anthracycline antineoplastic agent that intercalates DNA, inhibiting topoisomerase II, and inducing cell death in rapidly dividing cells. This combination regimen seeks to leverage multiple mechanisms of action to target leukemic cells in AML, particularly those harboring FLT3 mutations.
