Overview
**crisantaspase + CALGB regimen** is a combination therapy used in the treatment of acute lymphoblastic leukemia (ALL), particularly in adolescents and young adults (AYA). This regimen substitutes crisantaspase recombinant (Erwinia asparaginase) for pegylated asparaginase within the pediatric-inspired CALGB 10403 chemotherapy protocol, which includes multi-agent chemotherapy with asparaginase as a key component. Crisantaspase is a recombinant Erwinia chrysanthemi-derived asparaginase that depletes circulating asparagine, inhibiting protein synthesis and leading to leukemic cell death. The CALGB regimen involves cycles of chemotherapy (vincristine, daunorubicin, cyclophosphamide, cytarabine, methotrexate, 6-mercaptopurine, dexamethasone, etoposide) plus CNS prophylaxis[1][2][4][5].
