Crisugabalin (HSK16149) is a novel gabapentinoid and a highly selective oral calcium channel α2δ subunit ligand developed by Haisco Pharmaceutical Group. It is a gamma-aminobutyric acid (GABA) analogue that exhibits higher affinity and slower dissociation from the α2δ-1 subunit compared to older agents like pregabalin, which may translate to enhanced analgesic efficacy and fewer central nervous system side effects. In a clinical trial led by Beijing Tiantan Hospital, crisugabalin is being evaluated as an adjunct to conventional therapy—comprising antiviral drugs, NSAIDs, and opioids—for the treatment of acute pain in patients with herpes zoster. This combination aims to optimize pain control during the acute phase of the infection and potentially mitigate the risk of developing chronic postherpetic neuralgia.
