CRS-207 + pembrolizumab is a combination immunotherapy investigational regimen that includes CRS-207, a live, attenuated double-deleted Listeria monocytogenes (LADD) engineered to express the tumor-associated antigen mesothelin, and pembrolizumab, a monoclonal antibody targeting the PD-1 receptor (anti–PD-1 checkpoint inhibitor)[1][2][3][4][5]. CRS-207 is designed to stimulate cell-mediated immunity directed at mesothelin-expressing tumor cells, while pembrolizumab blocks PD-1 to enhance T-cell anti-tumor responses. Preclinical data suggested possible synergistic effects, but clinical trials in malignant pleural mesothelioma, gastric, gastroesophageal junction, and esophageal adenocarcinomas did not demonstrate significant efficacy, and development of CRS-207 was discontinued due to lack of clinical benefit[2][3][5].
