CS1001 (sugemalimab) is a fully human, full-length immunoglobulin G4 (IgG4) monoclonal antibody that targets programmed death-ligand 1 (PD-L1). By binding to PD-L1, CS1001 blocks its interaction with the PD-1 receptor on T-cells, thereby preventing tumor cells from evading immune detection and destruction. This mechanism enhances the immune system's ability to attack cancer cells. CS1001 is developed by CStone Pharmaceuticals using the OmniRat/OMT transgenic animal platform and has shown efficacy in multiple solid tumors and lymphomas. In clinical trials, it is often combined with platinum-based chemotherapy agents such as fluorouracil and cisplatin for enhanced anti-tumor activity, particularly in non-small cell lung cancer (NSCLC)[2][3][4][5]. Fluorouracil is a pyrimidine analog antimetabolite that inhibits thymidylate synthase, disrupting DNA synthesis in rapidly dividing cells. Cisplatin is a platinum-based chemotherapeutic agent that forms DNA crosslinks leading to apoptosis of cancer cells.
