Drug intelligence / Profile preview
CS2009 + capecitabine + oxaliplatin is an investigational combination regimen consisting of CS2009, a tri-specific monoclonal antibody targeting PD-1, VEGFA, and CTLA-4, administered alongside the standard chemotherapy doublet capecitabine (an oral prodrug of 5-fluorouracil) and oxaliplatin (an intravenous platinum-based agent), collectively known as CAPOX or XELOX. Developed by CStone Pharmaceuticals, this combination is designed to leverage the synergistic effects of multi-pathway immune checkpoint blockade, anti-angiogenesis, and cytotoxic chemotherapy. CS2009 enhances T-cell activation and reverses exhaustion by blocking PD-1 and CTLA-4, while its anti-VEGFA component inhibits tumor angiogenesis and remodels the immunosuppressive tumor microenvironment. The addition of capecitabine and oxaliplatin induces immunogenic cell death and directly inhibits tumor cell proliferation. The regimen is currently being evaluated in a Phase I/II clinical trial (NCT06741644) for patients with advanced solid tumors, with a particular focus on first-line treatment of proficient mismatch repair/microsatellite stable (pMMR/MSS) metastatic colorectal cancer.
