CT-322 + irinotecan is an investigational combination therapy for cancer, specifically studied in recurrent glioblastoma multiforme. CT-322 (also known as pegdinetanib or Angiocept) is a PEGylated Adnectin—a novel class of targeted biologics derived from the 10th domain of human fibronectin—engineered to selectively bind and inhibit vascular endothelial growth factor receptor 2 (VEGFR-2), thereby blocking angiogenesis and tumor vascularization[5][6][9][10]. Irinotecan is a chemotherapeutic agent that inhibits topoisomerase I, leading to DNA damage and cell death. The rationale for combining these agents is that antiangiogenic blockade by CT-322 may enhance the cytotoxic effects of irinotecan by disrupting tumor blood supply while directly killing tumor cells[1][2]. This combination has been evaluated in phase II clinical trials for recurrent glioblastoma multiforme.
