This is an autologous cell-based cancer vaccine composed of CD34+ hematopoietic progenitor cell-derived Langerhans-type dendritic cells (LCs) that have been electroporated with messenger RNA (mRNA) encoding three tumor-associated antigens: CT7, MAGE-A3, and Wilms tumor 1 (WT1). The process involves collecting stem cells from the patient for transplantation and using a portion to generate dendritic cells in vitro. These dendritic cells are then loaded with the specified antigen-encoding mRNAs via electroporation. When administered subcutaneously as a vaccine after autologous stem cell transplantation (ASCT), these modified LCs present the encoded antigens to T-cells, aiming to stimulate robust antigen-specific CD4+ and CD8+ T-cell responses against multiple myeloma tumor cells expressing these antigens. The approach is designed to enhance immune surveillance and reduce relapse risk post-transplantation by broadening the anti-tumor immune response[2][3][5].
