Drug intelligence / Profile preview
CTX-10726 + CTX-471 is an investigational combination of two antibody therapeutics developed by Compass Therapeutics. CTX-10726 is a tetravalent bispecific antibody targeting programmed cell death protein 1 (PD-1) and vascular endothelial growth factor A (VEGF-A), designed to simultaneously block checkpoint signaling and tumor angiogenesis. It exhibits a unique mechanism of action where VEGF-A triggers the cleavage and removal of PD-1 receptors from T cells. CTX-471 is a fully human IgG4 monoclonal agonist antibody targeting CD137 (4-1BB), a costimulatory immune receptor on T and NK cells. In preclinical models of checkpoint-inhibitor-resistant tumors, the combination of CTX-10726 and CTX-471 has demonstrated enhanced anti-tumor activity, increased immune cell recruitment, and a more permissive tumor microenvironment compared to either agent alone.
