Curcumin + CD68 antibody (CC-CD68Ab) is an experimental antibody-drug conjugate (ADC) consisting of a curcumin derivative chemically linked to a monoclonal antibody targeting the CD68 antigen. CD68 is highly expressed on tumor-associated macrophages (TAMs) and microglia within the tumor microenvironment, particularly in glioblastoma multiforme (GBM). The therapeutic strategy utilizes the antibody to selectively deliver curcumin to these immune cells. Once internalized, curcumin induces a phenotypic shift in TAMs from an immunosuppressive, pro-tumor M2-like state to a pro-inflammatory, tumoricidal M1-like state. This reprogramming leads to the recruitment and activation of natural killer (NK) cells and the direct elimination of tumor cells. Developed primarily by researchers at the City University of New York, this compound has shown efficacy in preclinical models of glioblastoma and cervical cancer.
