Overview

Curcumin + prednisolone is a combination of two active compounds—curcumin, a polyphenolic compound derived from the spice turmeric (Curcuma longa), and prednisolone, a synthetic glucocorticoid corticosteroid. This combination has been investigated in both topical and systemic formulations for its potential synergistic anti-inflammatory effects. Curcumin exhibits anti-inflammatory, antioxidant, immunomodulatory, antiseptic, and anticarcinogenic properties but suffers from poor bioavailability when used alone. Prednisolone is widely used for its potent anti-inflammatory and immunosuppressive actions but can cause significant side effects with systemic use. The rationale behind combining these agents is to enhance therapeutic efficacy while potentially reducing the required steroid dose or mitigating steroid-associated adverse effects. Studies have explored this combination in various delivery systems—including human serum albumin-based nanoparticles for targeted delivery in rheumatoid arthritis models[1][4], as well as topical mixtures (e.g., curenext gel plus prednisolone) for oral lichen planus[2]. Mechanistically, curcumin may increase levels of anti-inflammatory cytokines such as IL-10 while prednisolone suppresses pro-inflammatory cytokines; together they may provide superior control of inflammatory diseases compared to either agent alone[1][4].
