Combination of **cusatuzumab**, a high-affinity anti-CD70 monoclonal antibody, and **venetoclax**, a selective BCL-2 inhibitor. This combination is under investigation for the treatment of acute myeloid leukemia (AML), particularly in patients ineligible for intensive chemotherapy. Cusatuzumab targets CD70 expressed on leukemia stem cells (LSCs), leading to their reduction via antibody-dependent cell-mediated cytotoxicity. Venetoclax inhibits BCL-2, promoting apoptosis in LSCs by suppressing oxidative phosphorylation. Combined, these agents work synergistically to eliminate AML LSCs more effectively than either alone. Preclinical data suggest venetoclax upregulates CD70 on LSCs, increasing their susceptibility to cusatuzumab. This combination is being evaluated mainly in combination with azacitidine, the standard of care in AML[1][2][5].
