Overview
Custirsen + docetaxel is an investigational combination therapy studied primarily for metastatic castration-resistant prostate cancer and advanced or metastatic non-small cell lung cancer. Custirsen is a second-generation antisense oligonucleotide that targets the mRNA of clusterin, a chaperone protein overexpressed in many cancers and associated with treatment resistance. By inhibiting clusterin production, custirsen aims to sensitize tumor cells to apoptosis and enhance the efficacy of chemotherapy. Docetaxel is a taxoid antineoplastic agent that stabilizes microtubules, preventing their depolymerization and thereby inhibiting cell division, leading to apoptosis in cancer cells. The combination has been evaluated in clinical trials (notably the SYNERGY trial) but did not show significant improvement in overall survival compared to standard regimens[1][2][4][5][6][8].
