Overview

CuTPyP + F68 (also written as CuTPyP/F68) is a research-stage, nanoparticle-based drug delivery platform and photosensitizer used in photodynamic therapy (PDT). It is a porphyrinic metal-organic framework (MOF) composed of a porphyrin ligand (tetra(4-pyridyl)porphyrin, TPyP) coordinated with copper ions (Cu²⁺) and stabilized with the biocompatible surfactant Pluronic F68. This system is designed to target tumor cells via the enhanced permeability and retention (EPR) effect. In preclinical studies, it has been loaded with therapeutic agents such as curcumol (CUR) to form CUR@CuTPyP/F68, combining PDT with curcumol-enhanced cancer immunotherapy, or doxorubicin (DOX) to form F68-DOX@CuTPyP. Upon laser irradiation, the porphyrinic framework generates singlet oxygen (¹O₂), which directly kills tumor cells and triggers immunogenic cell death, promoting the infiltration of CD4+ and CD8+ T cells and the release of pro-inflammatory cytokines (IFN-γ, IL-12, TNF-α).
