Overview

CV301 + M7824 + N-803 is an investigational combination immunotherapy regimen for advanced cancers, particularly metastatic or refractory/recurrent small bowel and colorectal cancers. The combination includes: *CV301*: A poxviral-based cancer vaccine targeting tumor-associated antigens CEA and MUC1, delivered as modified vaccinia Ankara (MVA)-BN-CV301 and Fowlpox (FPV)-CV301. It is designed to stimulate the immune system to recognize and attack cancer cells expressing these antigens. *M7824* (Bintrafusp alfa): A bifunctional fusion protein that simultaneously targets programmed death ligand 1 (PD-L1) and acts as a transforming growth factor-beta (TGF-beta) trap. By blocking PD-L1, it inhibits immune checkpoint pathways that suppress T-cell activity; by trapping TGF-beta, it reduces immunosuppressive signaling in the tumor microenvironment. *N-803* (Anktiva; nogapendekin alfa inbakicept-pmln): An interleukin‑15 receptor agonist that promotes proliferation and activation of natural killer cells and CD8+ T cells, enhancing anti-tumor immunity. This combination aims to synergistically activate the immune system against tumors by combining antigen-specific vaccination with checkpoint inhibition/TGF-beta blockade and cytokine-driven lymphocyte activation[1][2][3][6].
