CV8102 + anti-PD-1 is a combination immunotherapy regimen under clinical investigation for the treatment of advanced solid tumors, particularly melanoma, cutaneous squamous cell carcinoma, head and neck squamous cell carcinoma, and adenoid cystic carcinoma. CV8102 is a synthetic single-stranded, non-coding RNA complexed with a cationic peptide that acts as an agonist of toll-like receptor 7/8 (TLR7/8) and retinoic acid-inducible gene I (RIG-I), thereby activating both innate and adaptive immune responses within the tumor microenvironment. When administered intratumorally, CV8102 stimulates local immune activation and enhances infiltration of T cells into tumors. The addition of systemic anti-programmed death protein 1 (PD-1) antibody therapy further boosts antitumor immunity by blocking PD-1-mediated inhibition of T cells. Preclinical studies have shown synergistic effects between these agents, leading to improved tumor control compared to either agent alone. Early-phase clinical trials indicate that this combination is generally well tolerated with preliminary evidence of efficacy in patients who are refractory or naïve to prior PD-1 inhibitor therapy[3][5][4][6].
