Overview

CV8102 + rabies vaccine is a **combination therapy** that pairs CV8102, a synthetic RNA-based immunostimulatory adjuvant, with a licensed rabies vaccine (Rabipur). CV8102 acts as a **toll-like receptor 7/8 (TLR7/8)** and **retinoic acid-inducible gene I (RIG-I)** agonist, designed to trigger innate immune responses and enhance adaptive immunity. When combined with the rabies vaccine, CV8102 significantly increases the immunogenicity of the vaccine, resulting in improved titers of rabies virus-neutralizing antibodies in healthy adults. The combination is administered intramuscularly and has been shown to improve the kinetics and magnitude of the antibody response alongside a favorable safety profile, though higher doses may cause transient flu-like symptoms or increases in markers of inflammation. The developer is CureVac, and the combination is primarily indicated for rabies prophylaxis and currently under investigation as a model for RNA-based adjuvant strategies.
