CVHNLC + pembrolizumab is an investigational combination therapy for advanced squamous non-small cell lung cancer (sqNSCLC), now in Phase 1 clinical trials. The therapy consists of CVHNLC, a novel mRNA-based precision immunotherapy developed by CureVac, and pembrolizumab, a programmed cell death protein 1 (PD-1) immune checkpoint inhibitor. CVHNLC comprises two different mRNA constructs encoding eight tumor-associated antigens, including both well-known and proprietary, novel solid tumor antigens identified through a whole genome-based platform. CVHNLC is formulated in lipid nanoparticles and is designed to foster targeted anti-tumor immune responses. When paired with pembrolizumab checkpoint inhibition, the combination is intended to amplify and prolong an immune-mediated attack on cancer cells. This therapy is being tested for safety and tolerability as first-line maintenance after chemotherapy (with pembrolizumab or pembrolizumab monotherapy) and in combination with first-line chemotherapy, specifically targeting metastatic sqNSCLC[1][3].
