**CX-072 + vemurafenib** is a combination therapy being investigated for the treatment of advanced or recurrent solid tumors and lymphomas. CX-072 is a Probody® therapeutic monoclonal antibody designed to target and inhibit programmed cell death ligand 1 (PD-L1), an immune checkpoint protein that tumors use to evade immune detection. This Probody is “masked” and only activated by tumor-associated proteases, aiming to minimize immune-related toxicity. Vemurafenib is a small molecule BRAF inhibitor that selectively inhibits the activity of mutated BRAF V600, a driver mutation found in several cancers, most notably metastatic melanoma. The rationale for the combination is to achieve synergistic anti-tumor activity via immune checkpoint blockade and direct inhibition of tumor cell proliferation. The combination is investigational and has been evaluated in early-phase clinical trials, notably the PROCLAIM-CX-072 study, sponsored by CytomX Therapeutics[1][3][5].
