Overview

CX-801 + pembrolizumab is an investigational combination therapy consisting of **CX-801**, a dually masked, conditionally activated interferon alpha-2b (IFNα2b) PROBODY cytokine developed by CytomX Therapeutics, and **pembrolizumab**, a monoclonal antibody PD-1 checkpoint inhibitor marketed as KEYTRUDA by Merck. CX-801 is designed to localize and activate the immune-stimulatory effects of interferon alpha-2b specifically in the tumor microenvironment, aiming to reduce systemic toxicities that have limited wider use of this cytokine in oncology. By combining with pembrolizumab, an anti-PD-1 immunotherapy, the regimen targets activation of the immune system via complementary mechanisms: direct enhancement of immune cell activity against tumors (via IFNα2b) and blocking of immune suppression (via PD-1/PD-L1 axis). The primary indication under investigation is metastatic melanoma, especially in PD-1 refractory patients. Phase 1 clinical evaluation is ongoing for advanced solid tumors, with dose escalation and expansion studies being conducted to establish safety, tolerability, and preliminary antitumor activity[1][2][3][4][5].
