Cyclo(prolylglycyl), also known as Traneurocin and by the code name NA-831, is a small molecule cyclic dipeptide belonging to the class of diketopiperazines. It is an endogenous neuroactive peptide found in human plasma and cerebrospinal fluid. Cyclo(prolylglycyl) acts as a modulator of AMPA receptors and has been shown to exert neuroprotective effects that are dependent on both AMPA receptor and TrkB receptor activation. It increases brain-derived neurotrophic factor (BDNF) in cell culture studies. The compound is under investigation for several neurological conditions including Alzheimer's disease, fragile X syndrome, Rett syndrome, major depressive disorder (MDD), and COVID-19-induced neuropathy. Its mechanism involves modulation of synaptic plasticity pathways relevant to cognition and neuroprotection[1][2][7].
