Cyclophosphamide + bortezomib + dexamethasone is a three-drug combination regimen used primarily as induction therapy for multiple myeloma. Bortezomib is a first-in-class proteasome inhibitor that blocks the 26S proteasome, leading to accumulation of proteins within the cell and subsequent apoptosis of malignant plasma cells. Cyclophosphamide is an alkylating agent (chemotherapy) that destroys rapidly dividing cells by cross-linking DNA strands and inhibiting DNA replication. Dexamethasone is a synthetic glucocorticoid steroid with anti-inflammatory and immunosuppressive properties; in this context, it helps kill myeloma cells and reduce inflammation associated with cancer or its treatment. This combination has demonstrated significant efficacy as both induction and consolidation therapy in newly diagnosed or relapsed/refractory multiple myeloma patients[1][2][3][4][6].
