This is a combination regimen of three small-molecule drugs—cyclophosphamide, celecoxib, and methotrexate—used in metronomic (continuous low-dose) chemotherapy protocols. Cyclophosphamide is an alkylating agent that crosslinks DNA, leading to cell death; methotrexate is an antimetabolite that inhibits dihydrofolate reductase, blocking DNA synthesis; and celecoxib is a selective cyclooxygenase-2 (COX-2) inhibitor with anti-inflammatory and potential antiangiogenic properties. The combination has been investigated primarily for its antiangiogenic effects in advanced cancers and relapsed or refractory lymphomas. The regimen aims to suppress tumor growth by targeting both cancer cells directly (cytotoxicity) and the tumor microenvironment (antiangiogenesis), with generally good tolerability at low doses[1][2].
