Overview

This is a combination regimen of three immunosuppressive drugs—cyclophosphamide, cyclosporin A (also known as ciclosporin or cyclosporine), and prednisone (sometimes substituted with prednisolone)—used primarily for the treatment of severe autoimmune conditions such as acute/subacute interstitial pneumonia associated with dermatomyositis or polymyositis. Cyclophosphamide is an alkylating agent that suppresses immune cell proliferation by cross-linking DNA. Cyclosporin A is a calcineurin inhibitor that blocks T-cell activation by inhibiting interleukin-2 transcription. Prednisone is a synthetic glucocorticoid corticosteroid that broadly suppresses inflammation and immune responses through multiple pathways including inhibition of cytokine production and leukocyte migration. This combination aims to provide intensified immunosuppression in life-threatening inflammatory lung disease where single-agent therapy has failed[1][6][8].
