Overview

This is an investigational four-drug combination regimen consisting of cyclophosphamide, doxorubicin, durvalumab, and nanoparticle albumin-bound paclitaxel (nab-paclitaxel). Cyclophosphamide is an alkylating agent that crosslinks DNA, leading to cell death. Doxorubicin is an anthracycline antibiotic that intercalates DNA and inhibits topoisomerase II. Nab-paclitaxel is a microtubule inhibitor formulated as albumin-bound nanoparticles to improve delivery and reduce hypersensitivity reactions. Durvalumab is a human monoclonal antibody targeting programmed death-ligand 1 (PD-L1), blocking its interaction with PD-1 and CD80 to enhance anti-tumor immune responses. This regimen has been studied primarily as neoadjuvant therapy in triple-negative breast cancer (TNBC), aiming to increase pathologic complete response rates compared to chemotherapy alone[1][3][6][8].
