Overview

This is a multi-agent combination chemotherapy regimen composed of nine drugs, each with distinct mechanisms of action. The agents include alkylating agents (cyclophosphamide, thiotepa), an anthracycline antibiotic (doxorubicin), antimetabolites (fluorouracil, methotrexate), a hormonal agent (tamoxifen), a synthetic androgen (fluoxymesterone), a vinca alkaloid (vinblastine), and a corticosteroid (prednisone). This combination targets cancer cells through multiple pathways—DNA crosslinking and damage, inhibition of DNA synthesis and repair, disruption of microtubule function during mitosis, modulation of hormone receptor signaling, and immunosuppression. Such regimens are designed to maximize antitumor efficacy by attacking cancer cells at different points in the cell cycle or via different biological processes. While several components are commonly used together for breast cancer treatment—such as cyclophosphamide, doxorubicin, fluorouracil ("FAC" regimen) often followed by tamoxifen—the inclusion of all nine drugs in one protocol is highly unusual and not standard clinical practice[1][2][3]. Each drug has established use in oncology; their combined use would be expected to result in significant toxicity.
